Is it a Vista OEM install ? Then removing unwanted startups and uninstalling programs / trialware would be one good way to start.

Remove crapware from your Windows Vista Computer may interest you.
Using the Registry Cleaner and a Registry Defragger would not result in any performance increase; however if you install/uninstall programs frequently, only then should you consider using one and that too, more from the point of good house-keeping.
CCleaner & Auslogics are safe and good freeware that I too use occasionally.
